All six Honey Sprite variants share the same base effect: spawning bees at the hit location when you deal ranged weapon damage. The variant you equip determines the additional effect you get on top of that.

There is no single guaranteed best location to farm the Honey Sprite since it can drop randomly anywhere across the map from any of those sources. That said, Shaken Sanctuary and Sunken Shores come up frequently for players hunting it, so they are worth prioritising if you want to increase your chances. The best consistent method is to open as many Pumpkin Chests as possible since they have a higher concentration of Sprite drops during Fortnitemares.
The Lucky Locator Gizmo is also worth using. It sits in your pickaxe slot at the start of every match and locates a buried Chest on the map that is guaranteed to contain a Sprite you have not collected yet. Once you have the Honey Sprite equipped, it activates automatically during matches. Every time you deal damage with a ranged weapon, there is a chance that bees will spawn at the hit location, dealing additional damage to your target. You do not need to activate it manually, and it does not take up an inventory slot beyond your Sprite slot.
The secondary effect of whichever variant you have equipped also activates automatically under its own conditions. The Trick or Treat variant triggers when you open containers, the Bounty Hunter variant triggers on eliminations; and so on. Stack the right variant with your playstyle, and the Honey Sprite adds a consistent passive advantage throughout a match.
